Salmonella spreads by targeting cells in our gut, study shows

Dec. 11, 2012 ? Scientists have gained fresh insights into how the salmonella bug makes us ill. University of Edinburgh researchers have found that the bacteria are able to change key cells that line the intestine, enabling the bugs to thrive.

By changing the make-up of these cells, the salmonella bacteria are able to cross the gut wall.

Food poisoning

Salmonella food poisoning -- commonly caused by eating undercooked poultry or eggs -- can lead to diarrhea, fever and even death in young children.

Scientists say the study furthers our understanding of how bacterial infections occur and what enables them to spread.

Cellular make-up

The University of Edinburgh research found that the salmonella released a protein -- SopB -- changing the make-up of certain cells that line the gut.

This causes a dramatic increase in cells -- called microfold or M cells.

The work, reveals how once the salmonella produces large number of these cells it can then get through into the bloodstream, causing infection.

"Bacteria have evolved sophisticated strategies to interact with and infect the host. This highlights yet another way in which microbes are able to transform cells into a type that suits their habitat," said Dr Arvind Mahajan, of The Roslin Institute at the University of Edinburgh.

The study was funded by the Biotechnology and Biological Sciences Research Council.

Egypt's journalists struggle under Mubarak-era laws

CAIRO ? A week before Egyptian President Mohammed Morsi issued a decree vastly expanding his powers and prohibiting judicial review of his decisions, an Egyptian satellite TV station, famous for helping re-energize the Egyptian uprising in February 2011, was forced off the air.

(Morsi, on Saturday, scaled back the decree, but this week's vote on a new constitution will go ahead as planned.)

The station, Dream TV, rose to international prominence after broadcasting an interview with former Google executive and Egyptian activist Wael Ghonim -- an interview that re-inspired the protest movement that led to the end of Hosni Mubarak?s presidency.?

On Nov. 15, the Egyptian government-run satellite company and broadcast authority cut the signal.? The minister of information, a member of the Muslim Brotherhood, said in a statement that the location of Dream TV?s studios violated the law because the studios are not in the Cairo suburb designated for live broadcasts.

Dream TV journalists disagree. They say the closure was a result of the station's criticism of Morsi?s government. ?The decision is not legal but political,? Dream TV anchor Wael Al-Ibrashy was reported as saying on air before the transmission was cut. ?The channel is being punished for a political stance.?

NBC tried to contact Information Minister Salah Abdel Maqsoud for comment but has not received a response.?

As protests continued, President Mohammed Morsi rescinded a controversial decree he issued weeks ago, assuming control of the judiciary. NBC's Ayman Mohyeldin reports.

Dream TV has since gone back on the air temporarily; it may be shut down again pending a court ruling.

The station?s struggle is just one of hundreds of battles being waged in the fight for an independent Egyptian media.?

Journalists in Egypt face around 30 Mubarak-era laws in the criminal code meant to muzzle the media, with the potential to punish journalists with hefty fines and years in jail.?

Hundreds of complaints
So far this year, Egyptian judges have filed at least 900 formal complaints against television channels and journalists, according to the English-language Al Ahram Online.?

?The laws haven?t changed,? said Cairo-based Human Rights Watch researcher Heba Morayef. ?The penal code criminalizes everything from insulting the president to insulting public institutions to insulting a foreign king.?

In early December, 11 Egyptian newspapers participated in a ?news blackout? to protest Morsi?s decree and potential constitutional restrictions on journalists.

Despite the government?s arrest and prosecution of journalists, the state?s actions have not had a chilling effect on speech. Egyptians can speak their minds more than at any time in most people?s memory, and there are dozens of new independent TV channels and newspapers.

?People are hungry for politics, and there?s a lot of new media,? said Said Sadek , an Egyptian political sociologist. ?It?s difficult today to restrict ideas.???

Even under Mubarak?s dictatorship, independent, privately owned media, like Dream TV, gave a platform to opposition and human rights groups -- within limits.

By contrast, the state-owned newspapers and TV stations were essentially Mubarak?s propaganda mouthpieces.

Although Egyptians favor independent, privately owned newspapers to the state-owned press, the state-owned, nationally broadcast TV channels are still very powerful. They are directly controlled by the government?s Ministry of Information. ?

Many journalists and free-speech advocates hoped that when Morsi took office and ended military rule in August, he would abolish the country?s Byzantine press rules and dismantle the Ministry of Information.

Instead, Morsi appointed Maqsoud, the fellow Muslim Brotherhood member, to run the ministry. Analyst Elijah Zerwan says state media?s coverage is more balanced than under Mubarak, but it still favors those in charge, as evidenced by recent coverage of the anti-Morsi protests in Tahrir Square and around Egypt.

?They are interviewing people in the square and showing protests, but also featuring a lot of guests from the Brotherhood,? Zerwan said.? ?So they seem to be struggling with it.?

That struggle appears to have pushed the head of state TV over the edge.? On Thursday Dec. 6, he submitted his resignation.

?I resigned in protest at the general administration of this regime,? former state TV head Essam Al-Amir said, as reported by the English-language Daily News Egypt.? ?It is not only about the?Egyptian state media. The entire political situation has pushed me to quit.?

Al-Amir said the government had requested that state TV programming include more voices that presented the government?s view, the Daily News reported.?

ANALYSIS: Egyptians warn Morsi is no friend of US

Journalist Rania Al Malky worries that Morsi?s government will take Egyptian media backward. ??

"There is this fear that it's going to be a return to how it was before, with the people in power controlling the message and framing the messages that are coming out of the national press,? she said.

Malky and other journalists fear Egypt?s new draft constitution enshrines the country?s restrictive press laws, notably by curbs on freedom of expression. At press time, the vote on the draft is scheduled for Dec. 15.

As large organisms, trees face some remarkable challenges, particularly regarding long-distance transport and communication. In addition to moving water and nutrients from their roots to their leaves, they must also integrate cell-to-cell communication over large areas. Furthermore, in order to function as a single, cohesive organism they must be able to effectively and efficiently send vital substances?such as DNA regulating signals?long distances along a network of cells, sieve-tubes, and vessels.

But how effective is this cell-to-cell communication and how far can such solutes travel?

Xylem and phloem serve as the two main arterial systems within a plant, and substances travel along two main pathways. Apoplasmic transport, in the system of cell walls, is important for movement of water and mineral solutes. Symplasmic transport, in which openings in the cell walls allow the cytoplasm of cells to be connected, facilitates the transport of nutrients, small ions, hormones, and transcription factors along a series of cells. Although this intercellular transport system is crucial for regulating cell differentiation and subsequent tissue maturation, it is not known how extensive these symplasmic pathways are and whether they can effectively transport solutes long distances.

The fact that woody tissues?specifically secondary xylem?contain long-living parenchyma cells, in addition to the dead cells primarily responsible for water transport, led Katarzyna Soko?owska and Beata Zag?rska-Marek (University of Wroclaw, Poland) to investigate whether symplasmic pathways in these living cells might play an important role in transporting solutes from xylem to actively dividing tissues, such as the cambium. They've published their study in the most recent issue of the American Journal of Botany.

"Molecular transport of hormones and other signals in cell-to-cell communication is slow," notes Soko?owska. "Yet information is propagated quickly over large distances such as a tree height or width. Moreover, billions of cells building up a tree must quickly and efficiently communicate with one another. This is a necessity for maintaining this large organism's integrity throughout its development."

Soko?owska and Zag?rska-Marek, in order to determine whether they could visually trace the pathways of solutes as those solutes travel up the living cells of the xylem network, cut 1- to 3-year-old branches from maple (Acer pseudoplatanus) and poplar (Populus tremula x P. tremuloides) trees and immersed the cut ends in a solution containing one of several fluorescent dyes.

After leaving the branches in the dye solutions for 3 to 7 days (adequate time for the dye to move a substantial distance), the authors cut cross- and tangential sections of the branches and found that dye was present in the living cells of both the radial and axial xylem parenchyma and ray and fusiform cambial cells?clear evidence that symplasmic transport occurred in both the secondary xylem and the cambial tissues. Moreover, dye was not only clearly seen in the cytoplasm of the cells, but also in the cytoplasmic bridges spanning walls between adjacent cells, confirming intercellular movement.

"Through the fluorescent marker application, we have successfully been able to visualize the transportation routes of nutrients or signaling molecules, which are regulated by changing states of cell-to-cell conductivity in time and in space," said Soko?owska. "They can be compared to traffic lights on crossroads, which control the road traffic and highway networks, ensuring fast and smooth transport between distant places."

Moreover, by using different dyes, the authors were able to see differences between the apoplasmic and symplasmic pathways. Although the apoplasmic dye was present in the cell walls of the secondary xylem, it was not visible in the cambial primary cell walls. This, together with the presence of the symplasmic dye in the cambial cells and in axial and radial xylem parenchyma, indicates the importance of the symplasmic pathway for transporting substances from the living xylem cells to the actively dividing cambial cells, which are crucial for developing new cells and tissues.

One of the most surprising and interesting finds was that the dye was not evenly dispersed throughout the cells despite the regular arrangement of rays in the cambium.

"We found that when fluorescent tracers are loaded to tree branches and migrate freely in the system of living cells, the tracer distribution becomes uneven," commented Soko?owska. "The regions appear with a high and low tracer content in the cells and they alternate along the branches. The pattern emerges in cambium, an embryonic tissue responsible for thickening of a tree through making its secondary conductive tissues, such as wood."

Father up from the source of the dye, the branch regions of rays filled with dye alternated with regions of rays that did not contain the dye?both dye and non-dye rays were composed of living cells with vigorous cytoplasmic streaming. The authors suggest that this pattern could be the result of differences in regulating the intensity of movement and unloading of dye from xylem to cambial cells in different areas.

"Most importantly, this regulation at the cellular level has a potential, through the spatial shift of the cellular states, of propagating important information in the whole population of interconnected plant cells over long-distances?similar to the green wave of streetlights for driving cars," Soko?owska stated.

In addition, this long-distance symplasmic movement from the xylem into the cambial rays is of utmost importance because the cambial cells are zones of active cell production and the intensively dividing cells in this region require many nutrients.

"Nowadays we are getting closer to understanding how cell-to cell communication can be translated into higher levels of plant body organization," Soko?owska concludes. "Cambium is a plant tissue composed of immortal, ever-dividing cells, equivalent to stem cells in animals. The more we learn about the mechanisms governing their functions, the better perspectives we get for directing morphogenetic processes in plants, animals and possibly humans."

"The wood produced by cambium is used commercially," she adds. "So, knowledge about cambial function and its regulation is also important for future modification of wood production quality."

(AP) ? Mosquito control officials in the Florida Keys are waiting for the federal government to sign off on an experiment that would release hundreds of thousands of genetically modified mosquitoes to reduce the risk of dengue fever in the tourist town of Key West.

If approved by the Food and Drug Administration, it would be the first such experiment in the U.S. Some Key West residents worry, though, that not enough research has been done to determine the risks that releasing genetically modified mosquitoes might pose to the Keys' fragile ecosystem.

Officials are targeting the Aedes aegypti mosquitoes because they can spread dengue fever, a disease health officials thought had been eradicated in the U.S. until 93 cases originated in the Keys in 2009 and 2010.

The trial planned by mosquito control officials and the British company Oxitec would release non-biting male mosquitoes that have been genetically modified to pass along a birth defect that kill their progeny before reaching maturity. The idea is that they will mate with wild females and their children will die before reproducing. After a few generations, Key West's Aedes aegypti population would die off, reducing the dengue fever risk without using pesticides and at relatively a low cost, the proponents say. There is no vaccine for dengue fever.

"The science of it, I think, looks fine. It's straight from setting up experiments and collecting data," said Michael Doyle, pointing to research Oxitec has had published in peer-reviewed scientific journals. He inherited the project when he took the lead at the Florida Keys Mosquito Control District in mid-2011.

The district's website says the modified genes will disappear from the environment after the mosquitoes carrying it die, resulting in no permanent change to the wild mosquito population. The district also says that the mosquito species isn't native to the Keys, nor is it an integral food source for other animals.

Dengue fever is a viral disease that inflicts severe flu-like symptoms ? the joint pain is so severe its nickname is "breakbone fever." It isn't fatal but victims are then susceptible at subsequent exposures to dengue hemorrhagic fever, which can be.

"It's very uncomfortable. You ache all over, you have a terrible fever," said Joel Biddle, a Key West resident whose dengue fever symptoms lasted more than a week in 2009.

Biddle is among those concerned about the Key West trial. He worries the modified genetic material will somehow be passed to humans or the ecosystem, and he wants more research done. He and other Key West residents also chafe at the fact that the project was in the works long before it was made public late last year.

Only female mosquitoes bite, so the modified genetic material wouldn't be passed on to humans, Mosquito control and Oxitec officials said. They also say they're being transparent about their data and the trial.

Real estate agent Mila de Mier has collected more than 117,700 signatures on a petition she posted on Change.org against the trial. Most come from outside the Keys, which de Mier says shows that tourists don't support the mosquito control district.

"We are dependent here on our tourists, and people from all over the country have been sending the message," de Mier said.

A University of Florida professor who studies mosquito control said Oxitec's technology works and evidence from the company's experiments elsewhere show it can control mosquito populations, but it's not clear whether its methods are as effective at controlling the risk of disease transmission. Phil Lounibos of the Florida Medical Entomology Laboratory also said it would take repeated releases of modified mosquitoes for the program to work, and the public outcry against genetically modified organisms, even when it's irrational, may be insurmountable.

"The public resistance and the need to reach some agreement between mosquito control and the public, I see that as a very significant issue, outside of the (operating) costs, since this is not just a one-time thing," Lounibos said.

The Aedes aegypti has shown resistance to pesticides used to control other species, and is the most difficult for the district to manage. Common in the Southeast and the Caribbean, it lurks in standing water around homes and businesses and can breed in containers as small as bottle caps.

District inspectors go door-to-door to remove the standing water where they breed, a time-consuming task. The district spends roughly $1 million a year to suppress Aedes aegypti, 10 to 15 percent of the agency's budget, Doyle said.

"Unfortunately, control of Aedes aegypti is a never-ending job," said Larry Hriber, the mosquito control district's research director.

In the trial, thousands of male mosquitoes bred by Oxitec would be released in a handful of Key West blocks where the Aedes aegypti is known to breed; the number of mosquitoes in those neighborhoods would be measured against the numbers from similar blocks where no modified mosquitoes were released.

The state's agriculture department oversees the mosquito control district, and Doyle said he would not expect any challenge from the state if the FDA signed off on the trial. The mosquito control district wouldn't need any local permit for the trial, either, but officials held a public meeting earlier this year and have posted information on the agency's website.

Still, it could take years for the FDA to approve the trial.

There hasn't been a case of dengue fever in Key West since November 2010, but two other cases were reported elsewhere in South Florida this fall.

The mosquito trial proposed for Key West wouldn't be the first release of genetically modified insects in the U.S.

In 2009, the U.S. Department of Agriculture's Animal and Plant Health Inspection Service concluded that integrating genetically modified pink bollworms, bred by Oxitec to be sterile but more competitive in mating than regular bollworms, into the agency's plant pest control program was "the environmentally preferable alternative" to combat the cotton pest. The program was discontinued, however, after officials found that the genetically modified insects were not as hardy as pink bollworms sterilized through irradiation, and that their use would cause farmers to lose their organic certification.

Oxitec said the USDA's environmental assessment is one of several examples of proof that the trial's risks and methods are being independently evaluated. The company has trials in Brazil, the Cayman Islands and Malaysia, and it says it's gotten positive reviews from the latter two governments. It also cites its published research in peer-reviewed journals.

But Biddle, the onetime dengue patient, wants Oxitec to continue testing the modified mosquitoes outside the U.S.

The Brains Behind ?Bubala Please? Speak Out | Heeb

There?s an awesome new series on the interwebs that?s both Jewy, and gangsta. Bubala Please follows friends and gangsters Jaquaan and Luis as they teach us stuff about being Jewish in a way that is a little less suburban meek, and a little more street chic.

The series was created by Jacob S. Salamon and Jared F. Bauer. Jacob is a filmaker, designer, and entrepreneur who was founding member of the social software and analytics company Bazaarvoice before deciding to pursue his dream of working in the entertainment world. Jared is a screenwriter, filmaker, and independent producer who recently produced Max Landis? internet short The Death and Return of Superman which?starred Elijah Wood, Mandy Moore, Ron Howard, Elden Henson, Sarah Shahi, and Simon Pegg. While a a lot less thug than Bubala Please, their own story is pretty inspiring.

How did the two of you come together to create this series?

Jacob: We actually met in college. Jared was studying film, and I was studying business. We met in Astronomy class, of all places. I completely admired that Jared was following his dream, and I told him then, ?I need to pursue business for now, but one day I?ll join you in Hollywood and we can make movies together.? That was six years ago. Three months ago I moved to LA, and this was our first project together.

The concept was created by a great friend of mine, Josh Cohen. Josh called me from London a couple months ago and said, ?I have this idea: what if you had two LA gangsters who spoke Yiddish?? Jared and I knew it had to be done, so we collaborated with Josh to create Bubala Please as you see it.

Tell me a little bit about your Jewish backgrounds. Did you go through the Hebrew School grind, or were you raised more Jew-ish? And did you have over-bearing Bubbies?

Jacob: I actually grew up along the US-Mexico border in a town called Laredo, Texas. My hometown is 95% Hispanic, and nearly every one of my childhood friends was Latino. I often joke that I grew up as a minority within a minority. My grandparents were Holocaust survivors from Lithuania and Germany, who eventually settled on the border to establish their new lives in America. My grandfather actually helped establish Laredo?s synagogue and Jewish community in the 1950s, which is where I went to Hebrew school. I hated Hebrew school growing up ??I have memories of making my mom literally drag me there. But, as with anything, age gives you perspective. I?m so glad I had that experience.?As for my bubbie ??she?s a Yiddish-speaker and loves the show. But she criticized that Luis uses too much onion in the latke episode.

Jared: I grew up in a suburb outside of Houston where there?s not much of a Jewish community. My parents are both pureblood New York Jews so growing up in conservative Texas was a bit disorienting. All of my friends were gentiles, so Judaism had virtually no presence in my social life, yet I grew up in a household that was very culturally Jewish. I went to Hebrew school and had a Bar Mitzvah, but that?s pretty much where it stopped. I watch plenty of Woody Allen movies so I feel that sufficiently relieves the guilt of discontinuing my Jewish studies. Both of my bubbies live in New York so they don?t get the chance to be too overbearing.

The process of ?hitting? in the online community can be strange, even arbitrary. What elements did you want to have to ensure this would be a success? How do you feel about the reactions you?ve gotten so far?

Jacob: We had no idea if people would like Bubala Please. We figured that if it was a huge flop, at least we could use the show as our company?s Hanukkah card. But Jared and I were ambitious: we wanted people to see our show. The first thing we needed was to make sure the show was as shareable as possible. In that vein, we created a simple website, posted the video to YouTube, and developed identities on Facebook and Twitter to ensure there was never an excuse for the show not getting passed around.

I had so many doubts about whether people would like the show or not. The opinion I most cared about was my brother?s. He saw our rough cut over Thanksgiving and said, ?You nailed it.? Then I knew we?d made something good.

Jared: We?re very excited about the reactions we?ve gotten thus far. We have people around the world writing to us about how much they love the show, how much they?ve laughed watching it, etc. I can?t ask for more than that. Web shows are still in their infancy, and as such, I don?t think the model for success has been defined yet. Sure, there are sensations like Epic Meal Time that generate a lot of ad revenue and sponsorship, but the landscape is changing quickly. With mainstream American households quickly adopting technologies that allow them access YouTube on their TV, I think web content is around the corner from exploding. Hopefully Bubala Please will be there when it happens.

Your actors, Marcus Wayne and Rick Mancia, are fantastic. Had you worked with them before? How did you find them?

Jared: When we first decided to do the show, I told Jacob that I wanted to look for stand-up/sketch comedians from some of the local comedy venues. I had been producing here in LA for two years before Jacob arrived so I reached out to all the people I know in the comedy circuit to ask for references. We got a bunch of recommendations, some of which were extremely talented comedians. However, in the end, we met Marcus and Rick through an open casting call we posted on LA Casting networks. I was going for the wrong approach. The stand up comedians tended to play it a little too ?big,? whereas Marcus and Rick were versatile actors who could really inhabit the roles and make it believable. We?re very luck to have found them.

Did you have to teach your actors a little bit of Yiddish and tradition? Or were they familiar?

Jacob: Marcus and Rick learned quite a bit during rehearsals and on-set. We had to teach them about gefilte fish, matzo balls, latkes, the whole bit. Marcus actually thought the combination of apple sauce and sour cream on the latkes was disgusting ??you can kind of see it in his face during the episode.

Jared: Actually it was the texture that turned his stomach. You?ll notice he takes a very small bite. This has actually been one of the most rewarding elements of making Bubala Please. Sharing our cultural backgrounds has proven to be a kind of bonding experience, which is not only great because we?re learning about other cultures across the cast and crew, but because it builds a great director/actor dynamic.

Is this a series that you hope would appeal to the goyim as well?

Jacob: When we created the show, we certainly were catering to Jews. We thought Jewish 20-somethings would love it, the same way we do. But we didn?t expect there to be such a following with 45-65-year-old women!

Jared: Most of the goyim I have shown BP to have really liked it. They may not get all of the Yiddish references, but they grasp the basic conceit (LA gangbangers meet Boca Raton retirees). I?m hoping with later episodes we are able to reach a wider audiences while continuing to cater to our Jewish fans.

In your latest episode, I?m legitimately impressed you included the Shecheyanu! How long did it take for your actor, Marcus, to learn it?

Jared: Quite a while. It was kind of like a Rabbi giving a Bar Mitzvah student a recording of his Haftorah portion. Jacob quickly recorded a rough version of the rap in garage band and sent it to Marcus and Rick to memorize. When that didn?t work, we recoded a phonetic breakdown of the whole prayer. It sounded like an episode of The Electric Company. By the time we were in our composer?s studio, Marcus had it down.
